Abstract Scope |
Hospital-acquired bacterial infections (HAIs) continue to yield major global challenges to human health, and contemporary disinfection practices show limited efficacy resulting in unsatisfactory therapeutic outcomes. New strategies that offer non-specific and broad protection are urgently needed. We report a novel bioinspired silver-ceria nanozyme (AgCNP) synthesized at a Ce:Ag molar ratio of 5:1, and with a higher Ce3+ surface fraction. Under both liquid and dry conditions, AgCNP eradicated Pseudomonas aeruginosa, Staphylococcus aureus, and methicillin resistant S. aureus both in planktonic and biofilm form. Remarkably, eradication occurred after four rechallenges with a multispecies, and cumulative inoculation of 1032 CFU/mL. A key antibacterial mechanism includes the surface generation of hydrogen peroxide. Transcriptomic analyses revealed AgCNP abrogated respiration, and nutrient availability, leading to programmed cell death. Our results reveal the utility of AgCNP as a high efficacy nanomaterial for use against clinically relevant bacterial species and opens up a new approach for alleviating HAIs. |
