Abstract Scope Hyperthermia therapy is a method of heating the cancer cells locally in a short period of time to eliminate the tumor without influent normal cells. Near-infrared radiation (NIR) has high penetration efficiency in tissues which can be used as an ideal energy source. Cancer cells have a net of negative surface charges on their cell membrane due to the high level of glycolysis in their metabolism, thus, we developed Fe3O4@Cu2-xS nanoparticles which are rendered with positive charges by cationic polymers coating for bioelectricity-mediated targeting. On top of the positively charged surface, folic acid (FA) modification is added to achieve biomarker-based cell targeting. Besides, the Cu2-xS coated Fe3O4 in the cores provides a strong absorption in the NIR region which generate more energy and has higher photothermal efficiency in hyperthermia therapy. This dual-targeting design with Fe3O4@Cu2-xS core demonstrated a high targeting and photothermal efficiency for hyperthermia therapy.
