Abstract Scope Small extracellular vesicles (sEVs), nano-sized vesicles secreted by cells, contain diverse biomolecules suitable for early disease detection. This study introduces a novel method using three-dimensional (3D) nanofibrous substrates and silver nanoparticles (Ag NPs) for enhanced surface-enhanced Raman scattering (SERS) detection. The structural and chemical properties of the substrates were analyzed using scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), and atomic force microscopy (AFM). A significant advancement is the direct monolayer deposition of Ag NPs on nitrocellulose fibers, facilitating the development of paper-based biosensors for ultra-low analyte concentrations. The fabrication begins with creating a hydrophobic surface, followed by Ag deposition, forming discrete nanoparticles and numerous detection hotspots. The hydrophobic substrate enhances analyte concentration near hotspots post-evaporation, while the 3D structure leverages the Raman spectrometer’s field depth. This cost-effective, scalable platform could broadly apply to various biomarkers and analytes, offering promising capabilities for trace-level, label-free detection in diverse bio-component systems.
