Abstract Scope |
Breast-cancer has the propensity to metastasize to bone resulting in skeletal failures and death. Lack of effective therapeutics for bone-metastasized breast-cancer result from inefficacy of animal models and unavailability of patient samples. We have developed a unique testbed of bone metastasis using 3D nanoclay-based tissue-engineered scaffolds. The extract from Rhodiola-crenulate has been investigated for breast-cancer. However, its efficacy for bone-metastasized breast-cancer is unknown. Here, we evaluate three plant-extracts (R.Crenulata, V.Macrocarpon, and O.Vulgare) for cytotoxicity against bone-metastatic breast-cancer on the testbed. All three plant extracts reduce cell viability in bone-metastasized breast-cancer. An activation of the proapoptotic marker p53 was observed for both 2D and 3D bone metastatic cultures. Further, the bone-metastatic cultures showed higher expression of anti-apoptotic marker bcl-2 compared to 2D. Further, none of the plant extracts induced cytotoxicity in healthy bone cells. Overall, our results show that phytochemically enriched plant extracts could serve as new therapies for bone-metastatic breast-cancer. |
