Abstract Scope |
Overloading of the temporomandibular joint (TMJ) can disrupt oxygen gradients, generate oxidative stress, alter cell behavior, and properties of cartilage and subchondral bone. Load induced inflammation and oxidative stress can affect zinc (Zn) and sulfur (S) levels and respective species in cells and tissues. Our studies using X-ray fluorescence imaging and X-ray absorption near edge spectroscopy on an overloaded TMJ illustrated zone-specific Zn- and S-species. Oxidized glutathione (GSSG) was colocalized with sulfoxide and organic sulfate in low Zn articular zone. A reduced form of glutathione (GSH) with negligible Zn was observed in the differentiating zone. The subchondral zone illustrated higher zinc, and sulfoxide colocalized with inorganic sulfate. Increased texturing from X-ray diffraction pattern of the 002-lattice plane in Zn dominated regions was identified. Results will be discussed within the context of organic and inorganic precursors that aid in pathologic biomineralization commonly diagnosed as radiopaque regions in patients with TMJ osteoarthritis. |
