Abstract Scope |
Ocular degenerative diseases progress, in part, from accumulated oxidative stress in the posterior region that damage cells and tissues within the fundus. Application of redox-responsive polydopamine nanoparticles was investigated as a novel treatment for ocular degeneration related to oxidative stress. The nanoparticle platform was utilized for drug delivery of novel protein therapeutics for synergistic treatment of the reactive oxygen species and harmful inflammation that propagates degeneration. Nanoparticles were synthesized via self-polymerization and characterized by dynamic light scattering and ZetaPals analyzer, as well as explored for their therapeutic effectiveness in vitro. The biomaterials were spherical in morphology, with an average size of 307.5 ± 27.9 nm. They provided antioxidant relief to primary porcine retinal cells as measured by DCFDA assay after co-treatment of oxidative challenge and improved long term cell viability of challenged cells. Taken together, the protein therapeutic loaded polydopamine nanoparticles provide antioxidant and anti-inflammatory protection against ocular degeneration. |
