Abstract Scope |
Decellularized heart matrix (DHM) therapy is a tissue engineering strategy to repair cell death and tissue disorganization that follows ischemic heart injury. DHM is a tissue-derived hydrogel that shows promise as a vehicle for stem cell transplantation. Natural biomaterials such as DHM exhibit weak mechanical integrity. To address this limitation, we designed crosslinkable DHM for soft lithography patterning to organize donor cells. The formulations were achieved by chemically modifying DHM with methacryloyl functional groups (DHM-MA) to enable crosslinking via UV. We first characterized DHM-MA degree of methacrylation, crosslinking, and micropatterning integrity. Different formulations were evaluated by atomic force microscopy, spectroscopy, microscopy, and cell culture fidelity. We show modulation of stiffness with different formulations and UV time. Cells cultured on DHM-MA attach and remain viable. The current evidence demonstrates that minimum topographical features can be lowered with higher aspect ratios dependent on crosslinking density. |
