Abstract Scope |
Introduction: Acute myocardial ischemia results in a cascade of downstream inflammatory responses causing scar formation which further exacerbates myocardial functioning.
Hypothesis: Localized delivery of growth factors loaded cardiac patch onto the damaged heart will improve angiogenesis and improve cardiac function.
Methods and Results: In the current study, a composite nanofiber cardiac patch (CNFP) was fabricated by electrospinning gelatin and polycaprolactone (200 µm) and studding it with mesoporous polydopamine nanoparticles (200-250 nm) loaded with basic Fibroblast Growth Factor (bFGF). The CNFPs were tested for bFGF release in vitro. The in vivo efficacy of CNFPs was tested by suturing them epicaredially onto the infarcted hearts induced by left anterior descending (LAD) coronary artery ligation in mice. CNFPs significantly preserved the cardiac function by improving ejection fraction and strain, when compared to the control (untreated) group as assessed by echocardiography.
Conclusion: CNFPs loaded with bFGF promote localized angiogenesis and improve cardiac function. |
